It is possible to hide Saturdays and Sundays by creating a custom timeline:

This way you can create a timeline that includes only the workdays.
If you want to display "duration" that doesn't include Saturdays and Sundays than you will have to calculate it yourself and store it in the database as a custom field (and display it in the Duration header column).
